A civilization is made of citizens, so people obviously have to be fed. Improve your tiles which can generate food and build facilities like granary and water mill to improve the production. Cities without food will stop growing and will also produce fewer resources.
Your technological advance relies on your Science generation. The more Science per turn you make, the faster you will research new technologies. The gain of Culture will allow you to enact new policies for your nation. Depending on your choices, these policies will bring you bonuses for combat, Science generation, production, faith or other aspects. It is also important to keep generating Tourism at a good rate. Like in many other games, Gold is absolutely important in Civilization. It is important to keep the people happy, otherwise they will rebel. In Civilization V their mood was measured in Happiness and in Civilization VI in Loyalty. It can be gained by many different means, but the simplest way is to keep people in the cities fed and build entertainment facilities, such as stadiums, circuses, colosseums, etc. Happiness/Loyalty can also be improved by your social policies, units garrisoned inside cities, access to ocean or rivers, religion, and luxury resources explored or commercialized by you, like gems, silk, etc. Friendship with city-states also makes people happy.
When you invade an enemy city, it will generate a lot of unhappiness/unloyalty until you stabilize it, so pay a lot of attention while trying a domination victory.
If you lack Happiness in Civilization V, your military units will lose combat strength and rebels will join units similar to the barbarians. In Civilization VI a whole city can declare independence and keep sending forces to fight you.
So, really, you do want to keep your people happy.
